Título: Aspectos Ultraestruturais, Histoquímicos e Proteicos do intestino médio de Lutzomyia (Nyssomyia) umbratilis Ward e Fraiha, 1977.

Abstract: Sand fly Lutzomyia (Nyssomyia) umbratilis is the primary vector of Leishmania (Viannia) guyanensis in the Amazon region. Infection occurs when the blood supply of the female sandfly. Protozoa are then confined to the digestive tract of the insect. During several steps of their development, the flagellates are attached to the gut wall by flagellum. However, the parasite needs to escape of a number of barriers in the sand fly gut before a complete blood meal digestion. Peritrophic matrix (PM) secreted by blood-sucking insect is one of those barriers. In this context, imaging of different regions of the midgut and PM, determination of protein composition of the latter need to be thoroughly investigated. Females of L. umbratilis, were collected in a military training area (CIGS, Manaus-Itacoatiara, AM, BR) and fed experimentally in rats. The sandfly's midgut and PM were analysed, timeframe of the PM formation after the blood meal ingestion and its degradation were determined. Important morphological details of the L. umbratilis sand fly PM and of the midgut were studied by transmission (TEM) and scanning (SEM) electron microscopies, its chemical composition was analysed by histochemistry. Complete formation of the PM was observed in 24 h in the dissections. It has a gelatinous aspect and degrades in 72 h. The second PM is secreted after a second- new blood meal. A well-formed fibrillar network structure of the PM was observed by TEM after 24 h, as well as its continuous degradation in 72 h. The ecto-peritrophic face of the midgut was found to be a thick mass with several transversal bands, the latter may evidently be attributed to muscles of the abdominal region, and the PM demonstrated the presence of lumps. Histochemical analysis showed that main PM component is protein, followed by glycoproteins (with different carboxylated and sulfated glycans) and neutral glycoconjugates. The proteins separated from the of midgut and PM of L. umbratilis fed in an interval of 24h to 72h were investigated using the 2-D gel electrophoresis (2-DE) and sequenced by ESI-QToF mass spectrometry. Also, one has to consider that genome of Lutzomyia has not been sequenced up to date. Thus, it was interesting to find out that 60 proteins identified by 2-DE were not correlated with those already deposited in known databases (NCBI, UniProtKB, FlyBast), among the total of 81 protein expressed. In this study, 21 proteins classified as glycoproteins (5), proteoglycans (3), nucleoprotein (7) and chromoprotein (1) were identified, five of them with unknown function. Some of those relate to the matrix formation, resistance, adhesion, structure, maintenance and degradation. For the first time, the PM structure is described in detail for L. umbratilis species using a combination of electron microscopy (SEM and TEM) and histochemical analysis. Possible roles of the PM structural components and their importance for the development of Leishmania parasites are discussed.
Resumo: A espécie Lutzomyia (Nyssomyia) umbratilis é o principal vetor da Leishmania (Viannia) guyanensis na região amazônica. A infecção ocorre no momento da alimentação sanguínea da fêmea de flebotomíneo. Os protozoários então ficam confinados ao trato digestivo do inseto. Durante várias etapas do seu desenvolvimento, os flagelados ficam ligados à parede do intestino médio abdominal pelo flagelo. No entanto, o parasito precisa escapar de um número de obstáculos no intestino do flebotomíneo, antes da digestão completa do sangue. A matriz peritrófica (MP) secretada por inseto hematófago pode atuar como uma das barreiras. Neste contexto, a imagiologia de diferentes regiões do intestino médio e da MP, a determinação da composição proteica destas estruturas precisa ser cuidadosamente investigada. Fêmeas de L. umbratilis foram coletadas em uma área de treinamento militar (CIGS, Manaus-Itacoatiara, AM, BR) e alimentadas experimentalmente em ratos. O intestino médio e a MP foram analisados. O tempo da formação da MP após a ingestão de sangue e sua degradação foi determinado. Detalhes morfológicos importantes da MP e do intestino médio abdominal foram estudados por meio de microscopia eletrônica de transmissão (MET) e de varredura (MEV), a sua composição química foi analisada por histoquímica e eletroforese em gel bidimensional aliada a espectrometria de massas ESI-QTof. Nas dissecções a fresco foi observado que a MP se forma completamente em 24 h, possui um aspecto gelatinoso e se degrada em 72 h. Uma segunda MP é secretada após um novo repasto sanguíneo. A MP apresentou uma estrutura de rede fibrilar bem formada e foi observada por MET após 24 h, assim como a sua degradação contínua em 72 h. A face ecto-peritrófica do intestino médio foi observada e considerada uma massa espessa com várias bandas transversais, esta pode, evidentemente, ser atribuída aos músculos da região abdominal. Nesta face, a MP demonstrou a presença de grumos. As análises Histoquímicas revelou que o componente principal da MP é proteína, seguido por glicoproteínas (com diferentes glicanos carboxilados e sulfatados) e glicoconjugados neutros. Das proteínas isoladas do intestino médio e da MP de L. umbratilis, em um intervalo de 24 h a 72 h de digestão sanguínea, foram expressas 81 das quais 60 não se correlacionaram com as proteínas já depositadas em bancos de dados conhecidos (NCBI, UniProtKB , FlyBast) levando-se em consideração que o genoma do gênero Lutzomyia não foi sequenciado até à data deste estudo. Do total expresso, 21 proteínas foram identificadas e distribuídas nos grupos das glicoproteínas (5), proteoglicanos (3), nucleuproteínas (7), cromoproteínas (1), das quais, cinco, com função desconhecida. Algumas destas proteínas estão correlacionadas com a formação, resistência, adesão, estrutura, manutenção e degradação da MP. Pela primeira vez, a estrutura da MP é descrita em detalhes para a espécie L. umbratilis usando uma combinação de microscopia eletrônica (MET e MEV) e por análise histoquímica. Eventuais funções dos componentes estruturais da MP e sua importância para o desenvolvimento de parasitos Leishmania foram discutidas.
