Título: Ecofisiologia, morfo- anatomia e atividade antifúngica de Protium altsonii Sandwith (Burseraceae): um estudo da semente até a fase adulta

Abstract: The Amazon forest presents a great vegetable and, therefore, molecular diversity, which can be understood as a source of studies for the generation of products of scientific and technological interest. Protium altsonii Sandwith is native from Amazon. There are also representatives of economic importance in the state of Amapá, called as “white pitch”, which exudes a white resin, volatile, with a musky scent, used by the community of San Francisco (RDS). Most of the researches on Protium restrict to the description of its chemicals compounds, widely used by the industry. Ecophysiological and morphological studies are spare and, for that reason, there is no accurate information regarding the conservation status of these species yet. In this way, the aim of this study was investigating P. altsonii throughout the study of ecophysiological and morphological aspects, aiming to contribute for the knowledge of the autoecology of the specie and, in addition, the better use and control of the production of bioactive molecules with a pharmacological and/or industrial potential. There were collected fruits in the Iratapuru-Jari Reserve, where they were morphologically and anatomically described, besides the morphoanatomy of young and ripe leafs and the trunk. They were also germinated and monitored until the stage of seedling. There were determined the growth and the gain of biomass, the concentration of foliar macronutrients and micronutrients, chloroplasts content, chlorophyll fluorescence and foliar quantitative anatomy. Likewise, there were identified the chemical components of the essential oils by chromatography and conducted the phytochemical prospection of the extracts and oil, in addition to the antifungal tests. The fruits are globose to ovoid, with slightly acute apex and short stipitate, showing one and, more rarely, two stones per fruit. The seeds are non-albumenized, with tegumentary dormancy, embryonic immaturity and the presence of phenolic substances. They present low germinative percentage. In transversal cut in the abaxial region, the ripe seed of P. altsonii is formed by coating of seminal wrap with collapsed cells and fleshy cotyledonary parenchyma with secreting cavities and presence of oil. For the growth in height and diameter of the root crown, the highest values were observed in plants submitted to an intermediary level of irradiance, and the lowest values in plants submitted to low irradiance. The nitrogen foliar content varied from 14,5 a 15,9 g kg-1 between treatments, with the highest values observed in shadow plants, comparing to other treatments. Regarding the fluorescence variables of chlorophyll a, it was verified that when plants were submitted to a higher irradiance, they presented a decrease in the energy use efficiency, in the intermediary treatment and full sun, as an indicative of chronic photoinhibition. However, with the emission of new leaves adapted to the new environment of irradiance, and after ripe – what happened two months after the beginning of the experiment –, the plants exhibited recovery of their photochemical efficiency with anatomic and physiological features more suitable to the new conditions. The essential resin oils of nine P. altsonii samples led to the identification of 86 chemical compounds, representing 98-100% of essential oils components. The monoterpenes were prevailing in all samples, although the content of sesquiterpenes in some samples was around 38,05%. Besides, there are some important differences in the variation of chemical composition and the quantities of components according to the period registered. Therefore, the presence of various chemical substances, coupled with the variability of the chemical profile established for each type of collection. The evaluation of the fungitoxi effect was applied to the resin oil samples of P. altsonii and it was observed that oils 1 (PA profile), 2 (PB) and 5 (PE) presented a better fungitoxic activity on filamentous fungus A. ninger and F. oxysporum, with the formation of zones of inhibition over 1,0cm, some corresponding to almost 50% more activity than the positive control (miconazole nitrate). P. altsonii is a potential source of antifungal agents. This study allowed the integration of different areas of knowledge adding important results for Burseraceae and the Protium gender. The results obtained in this thesis through morphology, ecophysiology, phytochemistry and antifungal activity enable to affirm that P. altsonii is a specie which shows a great phenotypic plasticity in forest areas and a large potential for industrial purposes.
Resumo: A floresta amazonia apresenta uma grande diversidade vegetal, e consequentemente, molecular, que poderá ser entendida como fonte de estudos para geração de produtos de interesse cientifica e tecnológico. Protium altsonii Sandwith é nativa da Amazônia. Tem também representantes de importância econômica no estado do Amapá, denominados de “breu branco”, que exsuda uma resina branca, volátil, de odor almiscarado usado pela comunidade de São Francisco (RDS). A maioria das pesquisas sobre Protium restringem-se a descrição de seus compostos químicos, bastante utilizados pela indústria. Estudos ecofisiológicos e morfológicos são escassos e, por este motivo, ainda não se tem informações precisas á respeito do status de conservação dessas espécies. Dessa forma, o objetivo desse estudo foi investigar P. altsonii, mediante o estudo de aspectos eco e morfofisiológicos, visando contribuir para o conhecimento da autoecologia da espécie, além, do aproveitamento e o controle de produção de moléculas bioativas com potencial farmacológico e/ou industrial. Foram coletados frutos na Reserva Iratapuru- Jarí, onde foram descritos morfologicamente e anatomicamente, além da morfoanatomia das folhas jovens e maduras e do tronco. Foram também germinados e acompanhados até a fase de plântulas. Foi determinado o crescimento e ganho de biomassa, teores de macro e micronutrientes foliares, teores de cloroplastídios fluorescência da clorofila e anatomia quantitativa foliar. Também foram identificados os constituintes químicos dos óleos essenciais por cromatografia e realizado a prospecção fitoquímica dos extratos e óleo, além dos testes antifúngicos. Os frutos são globosos a ovoides, com ápice levemente agudo e estipitado curto, apresentam um e, mais raramente, dois pirênios por frutos. As sementes são exalbuminosas, com dormência tegumentar, imaturidade embrionária e presença de sustâncias fenólicas. Apresentam baixo percentual germinativo. Em corte transversal, na região abaxial, a semente madura de P. altsonii é formada por revestimento do envoltório seminal com células colapsadas e com parênquima cotiledonar carnoso com cavidades secretoras e presença de óleo. Para o crescimento em altura e diâmetro do coleto, os maiores valores foram observados nas plantas submetidas a um nível intermediário de irradiância e os menores valores nas plantas submetidas à baixa irradiância. Os teores foliares de nitrogênio variaram de 14,5 a 15,9 g kg-1 entre os tratamentos, com os maiores valores observados nas plantas de sombra, comparado com os demais tratamentos. Quanto às varíaveis de fluorêscencia da clorofila a verificou-se que as plantas quando submetidas a maior irradiância elas exibiram diminuição da eficiência no uso da energia, no tratamento intermediário e pleno sol, como indicativo de fotoinibição crônica. Contudo, com a emissão de novas folhas adaptadas ao novo ambiente de irradiância, e após maduras, o que ocorreu dois meses depois do início do experimento, as plantas exibiram recuperação da sua eficiência fotoquímica com características anatomicas e fisiológicas mais adequadas as novas condições. Os óleos essenciais da resina de nove amostras de P. altsonii conduziram a identificação de 86 compostos químicos, representando 98 - 100 % dos constituintes dos óleos essenciais. Os monoterpenos foram predominantes em todas as amostras, embora o conteúdo de sesquiterpenos em algumas amostras tenha sido em torno de 38,05 %. Além disso, diferenças importantes na variação da composição química e nas quantidades dos constituintes de acordo com o período registrado. Portanto, a presença de diversas sustâncias química, atrelada ao a variabilidade do perfil químico estabelecido para cada tipo de coleta. A avaliação do efeito fungitóxico foi aplicado às amostras dos óleos resina de P. altsonii e se observou que os óleos 1 (perfil PA), 2 (PB) e 5 (PE) apresentaram a melhor atividade fungitóxica frente aos fungos filamentosos A. ninger e F. oxysporum com formação de halos de inibição acima de 1,0 cm, alguns correspondendo a quase 50% de atividade a mais que o controle positivo (nitrato de miconazol). P. altsonii é uma potencial fonte de agentes antifúngicos. Essse estudo permitiu uma integração entre diferentes áreas do conhecimento, agregando resultados importates para Burseraceae e o gênero Protium. Protium. Os resultados obtidos nessa tese através da morfologia, ecofisiologia, fitoquímica e atividade antifúngica permitem dizer que P. altsonii é uma espécie que apresenta uma grande plasticidade fenotípica em áreas de florestas e um potencial enorme para fins industriais.
