Abstract: The Curimatidae family, order Characiformes, have a wide geographic distribution in Central and South America, being found in greater abundance in the Amazon River basin. Classical cytogenetic approaches to elucidate evolutionary aspects have been taken to the genus Potamorhina that occur in the Amazon region (Potamorhina latior, P. altamazonica and P. pristigaster) and have shown large interspecific variation with respect to the diploid number, bucking the trend of chromosomal stability in the macrostructure of Curimatidae species (2n = 54 chromosomes) family. Furthermore, the distribution of heterochromatin also appears to be involved in the process of diversification in the genre. However, chromosomal physical mapping and comparative analysis of repetitive DNA sequences are essential for understanding the genomic organization of the group. Thus, this study aimed to characterize through cytogenetics technics the three species of the genus Potamorhina found near Manaus, and understand the evolutionary mechanisms involved in the karyotype differentiation of these species. For this we sampled 19 P. latior, 17 P. altamazonica and 12 P. pristigaster from Iranduba and Manacapuru, in the Amazonas state of Brazil. The technics applied in theses samples were the conventional Giemsa staining, C-banding, silver nitrate impregnation and fluorescent in situ hybridization with DNA probes (5S rDNA, 18S rDNA, Rex 1,Rex 3, tropomyosin 1 and telomeric sequences). Analysis showed variations for both classical cytogenetic markers as for molecular markers. Diploid number of 54, 56 and 102 chromosomes were shown for P. pristigaster, P. latior and P. altamazonica, respectively. Heterochromatic regions were present in the centromeric regions of chromosomes and also in the terminal regions. However P. latior provided further interstitial markings. The 5S rDNA and 18S were the simple type, not syntenic in Amazonian species of Potamorhina. retroelement Rex 1 and 3 were present in the centromeric region of all chromosomes for the three species as well as tropomyosin 1, except for some chromosomes in P. altamazonica. Given this, comparing the information obtained herein to previously proposed phylogenetic data, that the terminals and centromeric regions rich in heterochromatin seem to be the most dynamic regions in the genome and involved in the evolution process of the genus Potamorhina.

Abordagens de citogenética clássica têm sido realizadas para as espécies do gênero Potamorhina que ocorrem na região amazônica (Potamorhina latior, P. altamazonica e P. pristigaster) e têm demonstrado grandes variações interespecíficas com relação ao número diploide, contrariando a tendência da estabilidade na macroestrutura cromossômica das espécies da família Curimatidae (2n = 54 cromossomos). Além disso, a distribuição da heterocromatina também parece estar envolvida no processo de diversificação no gênero. Entretanto, mapeamentos físicos cromossômicos e análises comparativas de sequências de DNA repetitivo são imprescindíveis para o entendimento da organização genômica do grupo. Diante disso, este estudo buscou caracterizar citogenomicamente as três espécies do gênero Potamorhina encontradas nas proximidades de Manaus - AM e compreender os mecanismos evolutivos envolvidos na diferenciação cariotípica das mesmas. Para isso foram amostradas 19 Potamorhina latior, 17 P. altamazonica e 12 P. pristigaster nos municípios de Iranduba - AM e Manacapuru - AM, as quais foram submetidas à coloração convencional com Giemsa, bandeamento C, impregnação com nitrato de prata e hibridização fluorescente in situ com sondas de DNAr 5S e 18S, Rex 1 e 3, tropomiosina 1 e sequências teloméricas. As análises demonstraram variações tanto para marcadores da citogenética clássica quanto para os marcadores moleculares. Número diploide igual a 54, 56 e 102 cromossomos foram evidenciados para P. pristigaster, P. latior e P. altamazonica, respectivamente. Blocos heterocromáticos estiveram presentes nas regiões centromérica e terminal dos cromossomos das três espécies, contudo P. latior apresentou marcações intersticiais adicionais. As marcações de DNAr 5S e 18S foram do tipo simples, não sintênico nas espécies amazônicas de Potamorhina. Os retroelementos Rex 1 e 3 estiveram presentes na região centromérica de todos os cromossomos para as três espécies, bem como o gene tropomiosina 1, com exceção de alguns pares cromossômicos em P. altamazonica. Diante disso, ao relacionar os dados aqui obtidos aos dados filogenéticos anteriormente propostos infere-se que as regiões terminais e, principalmente, centroméricas ricas em heterocromatina, parecem ser as mais dinâmicas do genoma e envolvidas na carioevolução de Potamorhina.
