Título: Caracterização citogenética e análise do perfil de metilação em Melipona seminigra merrillae e Melipona interrupta manaosensis (Hymenoptera, Apidae)

Resumo: Melipona is a genus represented by 74 valid species of stingless bees distributed in the tropical regions of the planet. Melipona’ speciesare the most wellcharacterized chromosomally and they are divided into groups with higher or lower heterochromatic content, which may present small differences in the distribution of heterochromatin. These bees have two castes, queens and workers, which are differentiated from each other during larval stage depending upon genetic and food components. Using classical and molecular cytogenetic techniques, this work aimed to obtain information about the composition and organization of chromatin, as well as to understand the epigenetic mechanisms involved in caste differentiation of two subspecies of Amazonian stingless bees Melipona seminigra merrillae and Melipona interrupta manaosensis. The chromosomal number of 2n = 18 and 22 and n = 9 and 11 were confirmed for M. interrupta manaosensis and M. seminigra merrillae, respectively in females (2n) and males (n). Both species were characterized as belonging to the group with high heterochromatic content and the mapping of nucleoli organelles and 18S were in agreement with the described in the literature. Microsatellite (GA)15 mapping showed distributed markers in eucromatic regions for both species, whereas the mapping with (CA)15 showed different patterns, which may be related to genome size in these species or the activity of transposable elements. Epigenetically, it was found that the methylation profile differs between workers and queens regarding the intensity of bands from genomic DNA treated with restriction enzymes, showing that workers have more methylated CpG sites than queens This difference may influence the expression of genes related to caste differentiation, which adds data for the advancement of studies of epigenetic mechanisms within the genus with emphasis on the distinction of castes.
