Abstract: Aedes aegypti has great epidemiological importance, being arbovirus vector, and has been showing resistance to synthetic insecticides. Studies have shown that dilapiol, present in the essential oil of Piper aduncum, and its derivatives, may be alternatives for the control of this mosquito. In this work, methyl dilaphol ether (EMD) and temephos (TF) were compared for ovicidal and larvicidal activity, for four successive generations (G1 to G4), for genotoxic action and changes in A. aegypti development. The immature forms (eggs and larvae 3rd stage of A. aegypti) were exposed to concentrations of 60, 80, 100, 120 and 140 μg / mL of EMD, 0.002, 0.005, 0.007, 0.01 and 0.02 μg / mL of temephos and (water and DMSO 0.05%) for 24 and 48 hours to evaluate ovicidal and larvicidal activity and to determine lethal concentrations (LC50 and CL90). Pregnant adult females of A. aegypti, treated in the larval phase for 4 hours with EMD at 60.80 and 100 μg / mL and TF at 0.002, 0.005 and 0.007 μg / mL, were separated to estimate the oviposition rate for four generations (G1 to G4). The hatching rate was verified and the morphometry of the total egg length was performed. In each generation, cytological preparations of larval neuroblasts and oocytes were made to determine genotoxic action. After 24 h, EMD and TF, at their highest concentrations, 140 μg / mL and 0.02 μg / mL), caused 97% and 84% of egg infeasibility, respectively. The IC 50 and IC 90. were 60 and 119 μg / mL for the EMD, and 0.002 and 0.011 μg / mL for the TF. After 24 h, they caused 99% and 85% mortality of the larvae at the highest concentrations, and 100% mortality after 48 h at the concentration 120 and 140 μg / mL EMD and 0.02 μg / mL temephos. The LC50 and CL90 after 24 h with EMD were 97 and 140 μg / mL and in temephos, 0.005 and 0.019 μg / mL respectively, after 48 h it was 75 and 99 μg / mL in EMD, and 0.003 e 0.007 μg / mL in TF, respectively. Females treated in the larval phase with EMD and temephos, significantly reduced oviposition (Tukey, p <0.05), compared to the negative control, over the four generations. Both products reduced the hatching rate of treated female eggs relative to those in the control group. There was a significant reduction (p <0.05) in mean egg length of females treated with EMD and temephos in relation to the negative control, except the G1 and G4 generations. The genotoxic effect of EMD and TF on neuroblasts of larvae and oocytes of adults of A. aegypti, from G1 to G4, was recorded by means of the micronucleus, malformation, polynucleotide, nuclear bridge, budding and chromosome breaks, but, the EMD presented a higher frequency of these damages in G4, compared to the TF. The EMD, at the concentrations tested, presented higher toxic, genotoxic and developmental changes in A. aegypti compared to TF, but later studies will be necessary to better understand the effects of the EMD molecule.

Estudos têm mostrado que o dilapiol, presente no óleo essencial da Piper aduncum, e seus derivados, podem ser alternativas para o controle desse mosquito. Neste trabalho, o éter metil dilapiol (EMD) e o temefós (TF), foram comparados quanto à atividade ovicida e larvicida, por quatro gerações (G1 a G4) sucessivas, quanto à ação genotóxica e alterações no desenvolvimento de A. aegypti. As formas imaturas (ovos e larvas 3º estádio de A. aegypti), foram expostos às concentrações 60, 80, 100, 120 e 140 µg/mL do EMD, 0.002, 0.005, 0.007, 0.01 e 0.02 µg/mL do temefós e ao controle negativo (água e DMSO 0.05%), por 24 e 48 horas, para avaliar a atividade ovicida e larvicida e determinar as concentrações letais (CL50 e CL90). As fêmeas adultas grávidas de A. aegypti, tratadas na fase larval por 4 horas com EMD a 60, 80 e 100µg/mL e TF a 0.002, 0.005 e 0.007 µg/mL, foram separadas para estimar a taxa de oviposição, durante quatro gerações (G1 a G4). Foi verificada a taxa de eclosão e realizada a morfometria do comprimento total dos ovos. Em cada geração, fêz-se preparações citológicas, de neuroblastos de larvas e de ovócitos, para determinar ação genotóxica. Após 24 hs, o EMD e TF, em suas maiores concentrações, 140 µg/mL e 0.02 µg/mL), provocaram 97% e 84% de inviabilidade dos ovos, respectivamente. As CI50 e CI90, foram 60 e 119 µg/mL para o EMD, e de 0.002 e 0.011 µg/mL para o TF. Após 24 hs, estes provocaram 99% e 85% de mortalidade das larvas, nas maiores concentrações, e 100% de mortalidade após 48 h, na concentração 120 e 140 µg/mL do EMD e 0.02 µg/mL do temefós. A CL50 e CL90 após 24 hs, com o EMD foram, 97 e 140 µg/mL e em temefós, 0.005 e 0.019 µg/mL, respectivamente, após 48 h foi de 75 e 99 µg/mL em EMD, e de 0.003 e 0.007 µg/mL em TF, respectivamente. Fêmeas tratadas na fase larval com EMD e temefós, reduziram a oviposição significativamente (Tukey, p < 0.05), comparado ao controle negativo, ao longo das quatro gerações. Ambos produtos reduziram a taxa de eclosão de ovos de fêmeas tratadas em relação àquelas do grupo controle. Houve redução significativa (p < 0.05) no comprimento médio de ovos de fêmeas tratadas com EMD e temefós em relação ao controle negativo, exceto as gerações G1 e G4. O efeito genotóxico de EMD e TF em neuroblastos de larvas e ovócitos de adultos de A. aegypti, em G1 a G4, foi registrado por meio da ocorrência de micronúcleo, má-formação, célula polinucleada, ponte nuclear, brotamento e quebra cromossômica, mas, o EMD apresentou maior frequência desses danos na G4, comparado ao TF. O EMD, nas concentrações testadas, apresentou maiores efeitos tóxicos, genotóxicos e alterações no desenvolvimento em A. aegypti comparado ao TF, mas estudos posteriores serão necessários para maior conhecimento dos efeitos da molécula EMD.
